Gel permeation chromatography (GPC) is a type of size exclusion chromatography (SEC), that separates analytes on the basis of size.  It is often necessary to separate polymers, both to analyze them as well as to purify the desired product. GPC systems and consumables are now also available from a number of manufacturers.

EPRUI Biotech has developed monodisperse gel microspheres used for gel permeation separation of lipophilic polymers and hydrophobic molecules in the presence of organic solvents. The matrix of our gel permeation chromatography is styrene and divinylbenzne copolymer, which is nonporous transparent beads in dry form and will become porous network structure which has molecular sieve property after swelling in organic solvents. When the gel beads completely swell, the pore size or exclusion limits depend on the crosslinking level and eluant employed.

EPRUI GPC media are available with crosslink degree from 1-12% and have exclusion limits ranging from 400-14000 daltons.  Compared with other GPC media in the market, our monodispersed gel beads with precisely controlled diameter offer high separation resolution of lipophilic polymer and oligomers, low back pressure and minimal elution volume. The monodisperse polystyrene microspheres typically swell in benzene, toluene, xylene, carbon tetrachloride, cyclohexane, tetrahydrofuran, dimethylformamide, ketones, methylene chloride and other mixtures of solvents. Complete swelling is necessary before packing in the column and running the chromatography applications.

Applications

The GPC media can be widely used for the separation of small molecular weight organic compounds, such as pesticides, fungicides, polycyclic aromatic compounds and unsaturated lipids, but also widely used in agricultural pesticide residues in food and aflatoxin or Sudan Detection; polycyclic aromatic hydrocarbons, PCBs and organochlorine pesticides in water and soil.

EPRUI gel permeation chromatography media can be used for different eluent and different linear flow rates. The polystyrene beads with low cross-linking degree (1% and 3%) have stronger swelling ability and exclusion limit. However, the particles are soft and have poor pressure resistance, which is suitable for low linear velocity. While the gel beads with high crosslinking degree (8% and 12%) have higher mechanical strength, weaker swelling ability, lower exclusion limit, which can tolerate large linear velocity. Be sure to swell thoroughly before applying gel chromatography packing, and do not drip during application. Once the gel pack is dried, voids and faults are formed in the column, resulting in reduced separation and resolution.
